Audit your DNA sizing. Calculate fragment size (bp) based on migration distance relative to a molecular weight ladder.
DNA migration is inversely proportional to the log of its molecular weight. For accurate sizing, use a ladder where the sample falls within the linear range (middle of the gel) and maintain constant voltage/buffer concentration.
