Audit your cell concentrations. Calculate the cell density (cells/mL) based on hemocytometer grid counts and sample dilutions.
The standard hemocytometer volume is 0.1 mm³ per large square (10⁻⁴ mL). Accurate counting requires following the "border rule" (count cells on top/left lines, ignore bottom/right) to avoid double-counting.
